    基于LabVIEW的连续流动快速固相多肽合成

    Continuous flow rapid solid-phase peptide synthesis based on LabVIEW

    • 摘要: 为实现多肽固相合成的自动化和高效化,结合LabVIEW编程能力与流动合成技术,开发连续流动快速固相多肽自动合成系统,探索连续流动模式下单因素的优化策略。结果表明,相比于传统固相合成方法,该系统所需反应时间缩短近80%,试剂用量减少60%,兼具合成效率与经济优势;粗肽纯度高(阿基瑞林纯度达90.20%)。研究结论验证了该系统在短时间内合成高纯度多肽的能力,为多肽类药物开发及商业化生产提供可靠技术支撑。

       

      Abstract: In order to achieve automation and high efficiency in peptide solid-phase synthesis, a continuous-flow rapid solid-phase peptide synthesis system was developed. This system was developed by combining the programming capability of LabVIEW and the flow synthesis technology. In addition, the optimization strategy of single factor in continuous-flow mode was explored. The results demonstrated that, in comparison with the conventional solid-phase synthesis approach, the system shortened the reaction time by nearly 80% and decreased the reagents consumption by 60%, thereby exhibiting high synthetic efficiency and economic advantages. The crude peptide purity was found to be 90.20% for Argireline. The findings of this study demonstrated the capacity of the system to synthesize high-purity peptides in a brief period, thereby providing dependable technical support for the development and commercialization of peptide drugs.
